Lupron Depot Dosage for Central Precocious Puberty

The standard dose of Lupron Depot for central precocious puberty (early puberty in children) is based on the child's body weight, and will vary from child to child. The usual initial amount of Lupron Depot Pediatric is as follows:
  • Children who weigh 25 kg or less (55 pounds or less) will receive 7.5 mg every four weeks
  • Children who weigh 25 to 37.5 kg (55 to 82.5 pounds) will receive 11.25 mg every four weeks
  • Children who weigh more than 37.5 kg (82.5 pounds) will receive 15 mg every four weeks.
Your child's healthcare provider will monitor your child's response to treatment after one or two months and increase the amount if needed. The amount may be increased by 3.75 mg no more often than every four weeks until a dose is found that adequately stops puberty.
Two new once-every-three months strengths of Lupron Depot (pediatric) have been approved. These two strengths (11.25 mg and 30 mg) are given once every 12 weeks. No weight-based dosing guidelines are available for these products.
Once an adequate dosage is found, your child may still require periodic adjustments as he or she grows. Your child's healthcare provider will continue to monitor your child's response to determine if an adjustment is needed. The Lupron Depot injection should be stopped before age 11 in girls and age 12 in boys to allow for normal sexual maturation.

Dose of Lupron Depot for Endometriosis

Lupron Depot is available in one-month and three-month forms for endometriosis. The standard dosage will depend on the particular form used, as follows:
  • Lupron Depot 3.75 mg once a month
  • Lupron Depot-3 Month 11.25 mg once every three months.
For the initial treatment of endometriosis, Lupron Depot may be given alone or in combination with norethindrone acetate. The recommended length of treatment is six months.
If your symptoms recur after the initial six months, your healthcare provider may recommend a second six-month course of Lupron Depot. The second six-month course should always be given with norethindrone acetate 5 mg. Norethindrone helps reduce the risk of bone thinning that occurs when Lupron Depot is used long-term.
Lupron Depot-3 Month is not interchangeable with the monthly form of Lupron Depot. This means that Lupron Depot-3 Month 3.75 cannot be substituted for 3.75 mg of the Lupron Depot monthly form, regardless of how often the medications are given.
